While several recent publications in the field of clinical microbiology focus on PCR-based methodologies for the identification of microbial pathogens, the present manual describes mainly protein- based approaches for the diagnosis of infectious diseases. Rather than addressing specific pathogens, the book contains an exemplary collection of very detailed protocols that provide researchers, clinicians and students alike with established and novel methods for the identification of diagnostic markers, epitopes, and immunomodulatory components. Three of the eight parts of the book also deal with the heterologous expression, purification and structural characterization of diagnostically valuable microbial proteins. The latter are then covered to be used as diagnostic reagents in immunological assays. The book, designed to present a molecular approach to the identification of pathogens, could not entirely leave aside DNA-based methodologies, and those are aptly considered in additional chapters addressing representational difference analysis of cDNA derived from organisms whose genomes have not yet been completely sequenced, expression cloning aimed at producing diagnostically relevant proteins, and the analysis of the massive data generated by the use of DNA microarrays. The last chapter addresses problems of antibody engineering in providing details about the cloning of single-chain antibody fragments from hybridoma cells. This volume, written by about 60 experts from both academia and industry, is a useful reference source for any laboratory setting involved in the development or application of molecular diagnostic methods in medical microbiology.
