Comparative Genomic Hybridization (CGH) Detects Common and Distinct Aberrations in Non-Small Cell Lung Tumors of Different Histology

Aims: Molecular genetic and cytogenetic studies have revealed recurrent genomic aberrations in lung tumors. We performed CGH analysis on a series of non-small cell lung tumors of different histology to identify common and distinct genomic imbalances.

Methods: Surgical tissue samples were collected at the Faculty of Medicine of Uludag University. Genomic tumor DNA was isolated from fresh frozen tissue samples of 21 squamous cell carcinomas, 11 non- squamous cell lung carcinomas (6 adenocarcinomas, 4 large cell carcinomas, 1 adenosquamous carcinoma), and 1 atypical carcinoid. Tumor and normal reference genomic DNA were labeled by nick translation and hybridized to normal metaphase spreads. Fluorescense signal ratios from tumor and reference DNA were analyzed on a Vysis Quips Genetics Workstation.

Results: 17/21 squamous cell carcinomas, 9/11 non-squamous cell carcinomas and the atypical carcinoid revealed chromosomal imbalances. Overrepresentations were more frequent than losses (mean 2.9 vs. 1.5). Overrepresentations at distal 3q were more commonly found in squamous cell carcinomas than in non-squamous cell carcinomas (11/21 vs. 2/11). Similar trends were found for Overrepresentations at 18p (6/21 vs. 0/11) and 22q (5/21 vs. 0/11). Overrepresentations at 5p were detected in 3/21 squamous cell carcinomas and in 6/11 non-squamous cell carcinomas including all 4 large cell carcinomas.

Conclusions: The present data suggest common and distinct genetic pathways in non-small cell lung tumors of different histology. The observed imbalances may help to substantiate the events involved in the tumorigenesis of non-small cell lung tumors.
