Transcriptional Profile of Peripheral Blood CD34^Sup +^ Progenitor Cells From Polycythemia Vera Patients and Healthy Blood Donors

Aims: Polycythemia vera (PV) is a myeloproliferative stem cell disease characterized by an increased proliferation of all three myeloid lineages. The molecular pathogenesis of PV is unknown. We wanted to find out whether peripheral blood CD34^sup +^ progenitor cells demonstrate differential gene expression profiles compared to a healthy control group.
Methods: This study was performed on 5 patients using the RNA obtained from freshly separated peripheral blood CD34^sup +^ progenitor cells cultured for 12 hours in serum-free medium. Gene expression profiles were studied by hybridisation to cDNA array membranes that contain about 2,000 human genes (Clontech array technology). The results were compared with data obtained from healthy blood donors. Differential expression of a number of genes was confirmed using RT-PCR.
Results: We found that 10 genes were up-regulated >2.5 fold in all PV patients studied. Also some genes were down-regulated in the PV group. These genes are known to comprise anti-apoptotic and cell cycle pathways. They may represent a molecular signature for this disorder.
Conclusion: Specific expression changes are likely to be biologically important in the pathophysiology of PV.
